Ultrastructure of the Juxtaglomerular Cells in Streptozotocin-treated Rats


Abstract
This experiment was performed to study the effect of experimental diabetes on the ultra structural changes of the juxtaglomerular cells. Healthy adult male, rats weighing about 200 g each were used as experimental animals. Experimental diabetes, defined by, persistent hyperglycemia (>200 mg/100 ml) and destruction of pancreatic ¥â cells, was induced in rats by intravenous administration of 65 mg streptozotocin per kg of body weight. Diabetic rats were sacrificed under ether- anesthesia on the 30th day after the administration.
Renal and pancreatic specimens were fixed in formalin solution, embedded in paraffin, sectioned at a thickness of 4 am. Sections of kidney were stained by hematoxylin - eosin and PAS-, hematoxylin for general histologic findings. Sections of pancreas were stained by Gomori¢¥s aldehyde - fuchsin for the ¥â cells in the Langerhan¢¥s islets.
Renal tissues for electron microscopy were fixed in 2.5% glutaraldehyde in 0.1M phosphate buffer at pH 7.4. The tissue was postfixed in 1% osmium tetroxide solution. It was then dehydrated through graded ethyl alcohol and acetone, and embedded in Epon 812. The sections were cut with Sovall MT-2B ultramicrotome and stained with uranyl acetate and lead citrate. The observation was done with JEM-100CX electron microscope
The results were as follows:
1. Light microscopic findings revealed a mild increase cellularity and PAS. Positive mesangial matrix in the glomeruli of strepotozotocin-treated rats.
2. Electron microscopic findings of the juxtaglomerular cells following streptozotocin administration revealed poor development of the cell organelles, such as Golgi apparatus, granular endoplasmic reticulum, ribosomes and mitochondria. The specific granules of the juxtaglomer cells decreased in number and the granular matrix showed irregular electron density.
3. From these findings, it can be inferred that the juxtaglomerular cell activity, especially of the biosynthesis and release of the granules, decreases in streptozotocin-treated rats.
